What Is a Second-Source Standard?

A second-source standard is a calibration-verification material obtained from a source independent of the calibration standards or stock standards, when required by the analytical method or laboratory quality system.

The term “second source” describes the relationship between a verification material and the standards used for calibration. It is not a universal product type, and the exact terminology, source requirement, concentration, matrix, frequency, and acceptance criteria are method-specific.


Why Do Second-Source Standards Matter?

A second-source standard provides an independent check on the calibration system and can help reveal bias caused by a common source-material, dilution, matrix, transcription, or preparation error.

What Does “Independent Source” Mean?

An independent-source requirement means the verification material should not be traceable only to the same stock solution or preparation pathway used for calibration.

Depending on the method and quality system, source independence may involve:

  • A standard obtained from an outside commercial source
  • A material prepared from stock solutions different from those used for calibration
  • A different manufacturer or reference-material producer, when required
  • A separately prepared verification material with documentation supporting its source relationship
  • Certificate or lot documentation supporting traceability, concentration, matrix, uncertainty, and stability where applicable

IMPORTANT A different lot from the same supplier may or may not satisfy an independent-source requirement. The method, supplier documentation, and laboratory quality system determine whether the source relationship is acceptable.


Is a Second-Source Standard the Same as an ICV?

Not always. Initial Calibration Verification (ICV) describes a calibration-verification function. A second-source requirement describes the source relationship of the verification material.

Does an ICV Need to Be From a Second Source?

Only when the applicable analytical method, laboratory SOP, quality system, accreditation requirement, client requirement, or regulatory program requires an independent or second source.

Do not assume every Initial Calibration Verification (ICV) Standard, Continuing Calibration Verification (CCV) Standard, quality-control sample, or calibration-verification material has the same source requirement. Read the applicable method language first.


Is a Second-Source Standard the Same as a Continuing Calibration Check?

No. A second-source verification material checks calibration standards and/or instrument performance using an independent source when required. A continuing calibration check evaluates whether calibration remains acceptable during the analytical sequence.

The analytical function and the source relationship are separate questions. A continuing check may or may not have an independent-source requirement, depending on the method.


When Is an Independent Source Required?

An independent source is required when the analytical method, regulation, quality system, client requirement, or laboratory SOP requires it.

  • Initial verification of calibration standards or stock solutions
  • Periodic verification of calibration standards
  • Beginning use of a method
  • Preparation of new stock or calibration standard solutions
  • Corrective action after failed verification or suspected standard error
  • Laboratory quality or client-specific data-quality requirements

What Should Laboratories Check Before Selecting a Second-Source Standard?

Define the analytical requirement as precisely as possible before purchasing or preparing the verification material.

Identify the applicable method, terminology, source language, concentration, frequency, acceptance criteria, and corrective-action requirements.

How Should Source Independence Be Documented?

Document enough information to show how the verification material differs from the calibration source and to support review during audits or data evaluation.

Include the product name, catalog number, and lot number.
